| I. Background | |
| CLDN1 基因位于人类染色体 3q28.2，属于紧密连接蛋白家族（Claudins）成员。 CLDN1 基因编码的紧密连接蛋白 Claudin-1 为四次跨膜蛋白，包含两个细胞外环（ECL1、 ECL2）和 N/C 端胞内尾。ECL1 顺利获得二硫键稳定结构，参与细胞旁通道调控；C 端与胞内 支架蛋白结合，介导信号传导。 在上皮细胞和内皮细胞中，Claudin-1 顺利获得同型/异型相互作用形成紧密连接，调控细胞 旁通透性。而 Claudin-1 与胞内蛋白（如 Zonula Occludens-1）结合，参与 Wnt/β-catenin、 TGF-β等通路，影响细胞增殖、分化及迁移。在肿瘤中，其异常表达可激活促癌信号。 Claudin-1 作为紧密连接的核心组分，其功能贯穿屏障维持、信号调控及疾病发生。随 着对其机制的深入解析，针对 Claudin-1 的靶向治疗在肿瘤、慢性炎症及退行性疾病中展现 出广阔前景。 |
